Enteroviral 2C protein as a therapeutic target
Overview The poliovirus 2c protein is an RNA helicase that belongs to helicase superfamily 3 within a
larger AAA+ family of ATPases. When functional as a helicase, the superfamily 3 heilcases
(such as papillomavirus E1 that I studied as a postdoc) form a hexameric ring that encircles one
strand of nucleic acid. Thus far, poliovirus 2c has not been observed in a hexameric form,
indicating that a critical factor is missing to generate its functional form. The goal of this
subaward is to determine methods to biochemically prepare poliovirus 2c as a hexamer to
facilitate further biochemical and structural studies. This project will leverage my nearly 20
years of experience studying hexameric ring helicases that belong to the AAA+ family.
